His earliest memories were of pursuing a career in performing, having taken part in community theater, sketch comedy, and improvisation at an early age.
In 1998, Sestero moved to Los Angeles to pursue an acting career. He soon crossed paths with Tommy Wiseau, a mysterious figure claiming to be from New Orleans. In 2003, they together released the film The Room, a cult classic hailed as 'the Citizen Kane of bad movies'.

The Disaster Artist was an instant bestseller, and it has since earned Sestero a much larger following. In 2017, The Disaster Artist was adapted into an Academy Award-nominated film directed by James Franco and featuring an all-star cast. The film became a critical and commercial success, further cementing Sestero's place in Hollywood.
In 2016, Sestero wrote his second book, Best F(r)iends Volume One. The book follows the misadventures of its two protagonists: a businessman, Greg Sestero, and the mysterious drifter, Tommy Wiseau. It serves as a de facto pseudo-prequel to The Room, and it provides an insight into the lives of the two characters from their first meeting in 1998 until the filming of The Room. The book was released with an accompanying feature film and documentary, respectively titled Best F(r)iends Volume One and Best F(r)iends Volume Two.
Sestero currently splits his time between LA and New York, and is working on his third book.
